Members of the steroid hormone receptor family are important players in various cancers. Two of the most commons cancers affecting men and women, prostate and breast cancers, respectively, are addicted to hormone signaling (androgens and estrogens), making this class of proteins important drug targets. Drugs targeting hormone receptors have been the mainstay of cancer therapeutics for over three decades. However, resistance is inevitable and additional drugs with novel mechanisms of action are needed to manage the evolving cancer landscape. This special issue on novel next-generation steroid receptor ligands for the treatment of cancer will provide a compendium on evolving drugs targeting the nuclear hormone receptors and how the drug development process is adapting to treat aggressive resistant cancers.
